A Study on the Pulmonary Fibrosis in Rats after a Single Nonsurgical Intratracheal Dose of Bleomycin


Abstract
The purpose of this study was to develop the nonsurgical simple technique for intratraheal instillation in order to produce pulmonary lesions in the male rats. Sprague-Dawley male rats were divided into treatment group and control group.
The treatment group was given a single intratracheal dose of bleomycin hydrochloride(1.5 units/rat) and the control group was given a single intratracheal dose of normal saline(0.35§¢/rat).
The results were summerized as follows:
1. The survival rates of treatment group and control group were 75.0% and 88.9% respectively after dosing with the compounds.
2. The body weight gain of treatment group was decreased significantly in comparison with that of control group(p$lt;0.01). The lung wet weight of treatment group was increased significantly in comparison with that of control group(p$lt;0.01).
3. The rare(%) of lung wet weight to final body weight of treatment group was increased significantly in comparison with that of control group(p$lt;0.01).
4. The histopathological changes observed in the bleomycin-treated rats were atypia, dysplasia and hyperplasia of alveolar lining cells (Type II alveolar epithelium), Infiltrated mononuclear cells and macrophages into the alvclolar spaces. These changes were followed by accumulation of fibroblasts in the alveolar septa, and peribronchial and diffuse pulmonary fibrosis.
